    The Local Capacity, Local Community and Local Governance Dimensions of Sustainability in Australian Local Government

    The problem of the ‘financial sustainability’ of individual local councils represents the most significant policy question at issue in contemporary debate on Australian local government. This concern with financial sustainability has not only dominated almost all recent local government conferences across Australia, but it has also formed the capstone of several public inquiries into state local government systems. For instance, at the state level, both the South Australian Financial Sustainability Review Board’s (FSRB) (2005) Rising to the Challenge and the Independent Inquiry into the Financial Sustainability of NSW Local Government’ s (LGI) (2006) Are Councils Sustainable were centrally occupied with determining the meaning of financial sustainability in Australian local government and developing measures of financial sustainability. Moreover, the Queensland Local Government Association (LGAQ) (2006) Size, Shape and Sustainability (SSS) program, the Western Australian Local Government Association (WALGA) (2006) Systemic Sustainability Study and the Local Government Association of Tasmania (LGAT) (2007) Review of the Financial Sustainability of Local Government in Tasmania had at their core the problem of assessing financial sustainability in their respective local government systems

    An analysis of Singapore’s aware case

    Our case involves the women’s group Association of Women for Action and Research (AWARE), founded on 25 November 1985, which has proven itself to be a tireless long-term campaigner for women’s rights in Singapore. In this ‘AWARE case’, the executive committee of AWARE was surreptitiously taken over by fundamentalist Christian women from Church of Our Saviour at AWARE’s 28 March 2009 Annual General Meeting. In the second phase of this saga, the Extraordinary General Meeting on 2 May 2009 led to the secular ‘Old Guard’ recapturing AWARE and the Church of Our Saviour group being removed from power. This AWARE case shows the rise in power of fundamentalist Protestant Christianity within Singapore and its growing influence upon English-educated Chinese-Singaporeans. The fact that this upper middle-class demographic traditionally has supported the ruling People’s Action Party (PAP) is probably one key reason why the government decided not to intervene in this particular case as part of ‘the more consultative style of government’ first introduced by former PM Goh Chok Tong. The PAP government’s non-intervention in the takeover and recapture is extremely significant given its past record of authoritarian micro-management of dissent and oppositional opinions. It suggests more freedom for some NGOs in the contemporary era of Lee Hsien Loong’s prime-ministership. Significantly, PAP may be splitting into ‘reformist’ and ‘hardline’ factions headed by, respectively, Lee Hsien Loong and the MP for Bishan-Toa Payoh GRC Mr Wong Kan Seng

    The Cultural Ecology of Leadership: An Analysis of Popular Leadership Books

    Leadership is indisputably one of the most discussed, studied, and written-about topics in our society. A keyword search in the Expanded Academic Index for occurrences of the word leadership in a title or abstract reveals over 1,200 citations in the year 2000 alone. A subject search of leadership on Amazon.com returns more than 6,300 books on the subject, and over 1,400 hardcover books with leadership in the title are offered (Krohe, 2000). From Jesus CEO to 1001 Ways to Take Initiative at Work, fortunes are made (or not!) and fades are launched by many of these titles. But what wisdoms and lessons are truly to be gleaned from this popular genre of leadership writings? What techniques and approaches are most frequently utilized to deliver these so-called truisms? What can these leadership books tell us about how our society views the construct of leadership? And perhaps most importantly, how does this vast array of cultural knowledge about leadership and leadership processes affect leader-follower interactions? To answer these questions, we embarked on a qualitative and quantitative study of popular leadership books in order to understand this unique and fascinating genre

    Kaleidoscope JEIRP on Learning Patterns for the Design and Deployment of Mathematical Games: Final Report

    Project deliverable (D40.05.01-F)Over the last few years have witnessed a growing recognition of the educational potential of computer games. However, it is generally agreed that the process of designing and deploying TEL resources generally and games for mathematical learning specifically is a difficult task. The Kaleidoscope project, "Learning patterns for the design and deployment of mathematical games", aims to investigate this problem. We work from the premise that designing and deploying games for mathematical learning requires the assimilation and integration of deep knowledge from diverse domains of expertise including mathematics, games development, software engineering, learning and teaching. We promote the use of a design patterns approach to address this problem. This deliverable reports on the project by presenting both a connected account of the prior deliverables and also a detailed description of the methodology involved in producing those deliverables. In terms of conducting the future work which this report envisages, the setting out of our methodology is seen by us as very significant. The central deliverable includes reference to a large set of learning patterns for use by educators, researchers, practitioners, designers and software developers when designing and deploying TEL-based mathematical games. Our pattern language is suggested as an enabling tool for good practice, by facilitating pattern-specific communication and knowledge sharing between participants. We provide a set of trails as a "way-in" to using the learning pattern language. We report in this methodology how the project has enabled the synergistic collaboration of what started out as two distinct strands: design and deployment, even to the extent that it is now difficult to identify those strands within the processes and deliverables of the project.     Placing Joseph Banks in the North Pacific

    The South Pacific was a fulcrum of Joseph Banks's maritime world and global networks. The North Pacific was a distance and intangible fringe. This article is concerned with how Banks should be ‘placed’ in the North Pacific. It tracks how Banks's activities have been delineated in terms of languages and categories of global and local, and centre and margin, and then considers the historical and geographical specifics apposite to his connection to the North Pacific. In this setting, ideas of place (as location and assignment) and capital (as a circulatory and everyday practice of exchange and opportunism) come into view and question the distinction between science and commerce in Banks historiography. The article considers a diverse group of non-Indigenous figures – explorers, traders, cartographers, scientists, collectors – operating in the North Pacific in the 1780s and 1790s whose initiatives and missives passed across Banks's desk, and assesses their place in Banks's archive by drawing on Peter Sloterdijk's ideas about the interiorising and exteriorising logic of capital.PostprintPeer reviewe

    PAM variants in patients with thyrotrophinomas, cyclical Cushing’s disease and prolactinomas

    IntroductionGermline loss-of-function variants in PAM, encoding peptidylglycine α-amidating monooxygenase (PAM), were recently discovered to be enriched in conditions of pathological pituitary hypersecretion, specifically: somatotrophinoma, corticotrophinoma, and prolactinoma. PAM is the sole enzyme responsible for C-terminal amidation of peptides, and plays a role in the biosynthesis and regulation of multiple hormones, including proopiomelanocortin (POMC).MethodsWe performed exome sequencing of germline and tumour DNA from 29 individuals with functioning pituitary adenomas (12 prolactinomas, 10 thyrotrophinomas, 7 cyclical Cushing’s disease). An unfiltered analysis was undertaken of all PAM variants with population prevalence <5%.ResultsWe identified five coding, non-synonymous PAM variants of interest amongst seven individuals (six germline, one somatic). The five variants comprised four missense variants and one truncating variant, all heterozygous. Each variant had some evidence of pathogenicity based on population prevalence, conservation scores, in silico predictions and/or prior functional studies. The yield of predicted deleterious PAM variants was thus 7/29 (24%). The variants predominated in individuals with thyrotrophinomas (4/10, 40%) and cyclical Cushing’s disease (2/7, 29%), compared to prolactinomas (1/12, 8%).ConclusionThis is the second study to demonstrate a high yield of suspected loss-of-function, predominantly germline, PAM variants in individuals with pathological pituitary hypersecretion. We have extended the association with corticotrophinoma to include the specific clinical entity of cyclical Cushing’s disease and demonstrated a novel association between PAM variants and thyrotrophinoma. PAM variants might act as risk alleles for pituitary adenoma formation, with a possible genotype-phenotype relationship between truncating variants and altered temporal secretion of cortisol

    Phosphorylated Dihydroceramides from Common Human Bacteria Are Recovered in Human Tissues

    Novel phosphorylated dihydroceramide (PDHC) lipids produced by the periodontal pathogen Porphyromonas gingivalis include phosphoethanolamine (PE DHC) and phosphoglycerol dihydroceramides (PG DHC) lipids. These PDHC lipids mediate cellular effects through Toll-like receptor 2 (TLR2) including promotion of IL-6 secretion from dendritic cells and inhibition of osteoblast differentiation and function in vitro and in vivo. The PE DHC lipids also enhance (TLR2)-dependent murine experimental autoimmune encephalomyelitis (EAE), a model for multiple sclerosis. The unique non-mammalian structures of these lipids allows for their specific quantification in bacteria and human tissues using multiple reaction monitoring (MRM)-mass spectrometry (MS). Synthesis of these lipids by other common human bacteria and the presence of these lipids in human tissues have not yet been determined. We now report that synthesis of these lipids can be attributed to a small number of intestinal and oral organisms within the Bacteroides, Parabacteroides, Prevotella, Tannerella and Porphyromonas genera. Additionally, the PDHCs are not only present in gingival tissues, but are also present in human blood, vasculature tissues and brain. Finally, the distribution of these TLR2-activating lipids in human tissues varies with both the tissue site and disease status of the tissue suggesting a role for PDHCs in human disease

    Recruiting a New Substrate for Triacylglycerol Synthesis in Plants: The Monoacylglycerol Acyltransferase Pathway

    BACKGROUND: Monoacylglycerol acyltransferases (MGATs) are predominantly associated with lipid absorption and resynthesis in the animal intestine where they catalyse the first step in the monoacylglycerol (MAG) pathway by acylating MAG to form diacylglycerol (DAG). Typical plant triacylglycerol (TAG) biosynthesis routes such as the Kennedy pathway do not include an MGAT step. Rather, DAG and TAG are synthesised de novo from glycerol-3-phosphate (G-3-P) by a series of three subsequent acylation reactions although a complex interplay with membrane lipids exists. METHODOLOGY/PRINCIPAL FINDINGS: We demonstrate that heterologous expression of a mouse MGAT acyltransferase in Nicotiana benthamiana significantly increases TAG accumulation in vegetative tissues despite the low levels of endogenous MAG substrate available. In addition, DAG produced by this acyltransferase can serve as a substrate for both native and coexpressed diacylglycerol acyltransferases (DGAT). Finally, we show that the Arabidopsis thaliana GPAT4 acyltransferase can produce MAG in Saccharomyces cerevisiae using oleoyl-CoA as the acyl-donor. CONCLUSIONS/SIGNIFICANCE: This study demonstrates the concept of a new method of increasing oil content in vegetative tissues by using MAG as a substrate for TAG biosynthesis. Based on in vitro yeast assays and expression results in N. benthamiana, we propose that co-expression of a MAG synthesising enzyme such as A. thaliana GPAT4 and a MGAT or bifunctional M/DGAT can result in DAG and TAG synthesis from G-3-P via a route that is independent and complementary to the endogenous Kennedy pathway and other TAG synthesis routes
